In Borderlands 2, Three Horns Valley becomes available to us when we first travel to Three Horns Divide, but its not in a good idea to go there unless we acquire some optional missions from Sanctuary or until we complete Hunting the Firehawk story mission.

Borderlands 2 Cult Following: The Enkindling is the fourth and final mission in the Cult Following series that can be acquired by speaking to Incinerator Clayton, after we finish the previous mission, Cult Following: Lighting the Match.

Borderlands 2 Cult Following: Lighting the Match mission is granted to us by completing Cult Following: False Idols and speaking with Incinerator Clayton in Frostburn Canyon.

Borderlands 2 Cult Following: False Idols can be unlocked by completing the Cult Following: Eternal Flame mission we receive from Lilith.

After we save Lilith in Hunting the Firehawk mission we can travel back to Sanctuary and speak to her in order to get a new optional quest known as the Borderlands 2 Cult Following: Eternal Flame mission.

Borderlands 2 Frostburn Canyon is a smaller area connected to Three Horns Divide, and even if it is available when we first enter Three Horns Divide, its not in our interest to visit it before we have a mission that takes us here.

Another optional mission that becomes available to us after we complete the Plan B story mission is called Claptraps Secret Stash.

Borderlands 2 Do No Harm is an optional mission available after we complete the story mission called Plan B.

Borderlands 2 Rock, Paper, Genocide Corrosive Weapons is the third mission in the short Rock, Paper, Genocide series of missions we acquire from Marcus in Sanctuary.

Borderlands 2 Rock, Paper, Genocide Shock Weapons is another optional mission activated right after the Rock, Paper, Genocide: Fire Weapons! optional mission.
